Design and Biological Evaluation of Novel Imidazole Compounds

Prostate cancer is the most common cancer in men in the USA. Approximately 1.4 million cases are reported every year. Successful treatment varies depending on stage of diagnosis and aggressiveness of the cancer. Previously, our group reported that imidazopyridines have unique antiproliferative properties against castration-resistant prostate cancer cells. However, the efficacy of these compounds is limited by their solubility. The objective of this research is to develop new chemistry that can yield new compounds with enhanced properties against castration-resistant prostate cancer. Imidazole and Imidazopyridine-based small organic molecules are promising candidates for anticancer therapeutic research. Imidazopyridines exhibit antiproliferative properties against cancerous cells through a variety of molecular mechanisms such as vascular endothelial growth factor (VEGF)-receptor inhibition and KDR inhibition, which induces apoptosis. Two classes of nitrogen-containing heterocycle compounds have been prepared via a one pot synthesis using modified aromatic aldehydes and benzil. Salicylaldehyde and derivatives are modified and used to synthesize new imidazole and imidazopyridine compounds, designed to increase biological availability by adding hydrophilic groups. The newly synthesized compounds are characterized to confirm the desirable composition and structure. Biological evaluation is conducted via cytotoxic and migration studies. Results thus far have shown that functional group modifications to the imidazole compounds influence the inhibition of prostate cancer cell growth.
