Changes in acyl ghrelin, insulin and leptin of young obese Indonesians following a 4-weeks weight loss program

Background Weight loss improves acyl ghrelin (AG), insulin, leptin levels and is correlated with dietary intake reduction in population with obesity. Thus, diet plus exercise contribute to the dependent effect of appetite hormones. Thus, we investigated the effect of a 4-week weight loss program with diet and exercise on appetite hormone in young obese participants. Our hypothesis was weight loss program would result in decreased acyl ghrelin, and improved insulin and leptin levels. Methods Thirty-three participants (23 females, 10 males) with BMI >25 kg/m2, aged (19-20 years) were randomized into two groups: diet-exercise (n=18) and control group (n=15). The weight loss program was given for four weeks. Subjects received a hypocaloric diet containing high fiber, and aerobic exercise was twice a week. The goal for the obese was to lose 0.5 kg/wk by reducing their intake by 500 kcal/d. The biomarkers were assessed by enzyme-linked immunosorbent assay. Results The diet plus exercise group showed a significantly decrease in AG and insulin (-93.08±171.73 pg/mL; -9.72±14.75 ng/mL) compared to those (-15.55±27.49 pg/mL; -0.37±8.70 pg/mL) in CG (P<0.001; P<0.042). Furthermore, both groups revealed an increase of leptin (14.69±27.87 pg/mL for diet plus exercise group; 5.35±37.98 pg/mL for the control group. Conclusion These findings support diet plus exercise for weight loss in young obese through appetite hormones improvement
