Ischemic disease or coronary artery disease generate from that the heart is not getting enough blood flow and oxygen to supply the cardiac muscle (1). In a severe case, the cardiac muscle is dead (2). Currently, to use biomaterials as s cardiac patch scaffold is proposed to substitute the dead cardiac muscle (3). Therefore, to create the performance cardio patch scaffold is challenge for biomaterials scientists. In this research, polyurethane (PU) that has the suitable elastic properties for a cardio patch scaffold was created into nonwoven fibers by electro-spinning. Those nonwoven fibers were modified with reconstructed collagen assembly that could enhance biological functionality (4). To prepare electro-spun fibers, PU was dissolved in different solvents; MF, DMF:THF (75:15) and DMF: THF (50:50), respectively that the ratio of polyurethane and each solvent are 1:5 by weight. For electro-spinning process, the polymer solution was filled in the glass syringe and applied 10 kV for fiber fabrication. The distance between nozzle and collector is 8, 9, 10, and 11 cm, respectively. Then, to modify scaffolds, electro-spun fibers were immersed in collagen in 0.1 M acetic acid for 24 h before put in PBS, pH 7 for 24 h. Afterward, modified scaffolds were dried at the room temperature. To consider morphological structure and physical properties, scaffolds were observed by Scanning Electron Microscope (SEM) and Contact Angle Measurement. As the results, it founded that increasing distance between nozzle and collector decreased fiber diameter. The results of contact angle showed that small diameter fiber affected on decreasing of contact angle. For the modified scaffolds, the result showed that the fibrillar structure of reconstructed collagen assembly adhered on the electro-spun fibers. Finally, we expect that this modified scaffold promise to use for ischemic disease. However, to improve the biological functionaliy need for the future works.
