Systemic lupus erythematosus (SLE) is a systemic autoimmune disease of worldwide distribution, affecting predominantly women, characterized by presence of humoral and cellular abnormalities influenced by epigenetic and genetic factors. TLR7 and IRF-5, involved in the production of IFN-α, play a role in SLE. There are no genetic studies in the mayan population from Yucatan. Analysis of TLR7 rs179008 and IRF-5 rs2004640 were determined in 108 female patients and 200 healthy females. Frequency of T and A/T y T/T of rs179008 was lower in patients and no association was observed with SLE (OR 0.6527, OR 0.748 and 0.058, respectively). Frequency of T (56 vs 48%) and TT (33 vs 26%) of rs2004640 was higher in patients respect to controls (p = 0.037), Data suggest that rs2004640 is associated with the development of SLE in the mayan female population. Additionally, Expression of IRF5 (mRNA) was analyzed in patients and controls, observing significant increase in patients (p <0.0001). The results suggest the importance of rs2004640 in IRF5 expression and its role in SLE.
