In silico characterization of DNA motifs associated with the differential expression of the ornithine decarboxylase gene in the red macroalgae Grateloupia imbricata during in vitro cystocarp development

The formation of the reproductive structures, namely cystocarps, in the red seaweed Grateloupia imbricata is affected by diverse stimuli, both endogenous and exogenous; being the polyamines (PAs) and the gaseous compound, ethylene, the primary responsible for in vitro induction of the cystocarp. PAs synthesis is mainly catalyzed by the ornithine decarboxylase enzyme (ODC) which is regulated at the cellular level both by the proteasome and at transcriptional level. However, knowledge about gene regulation pathways in macroalgae, and the factors involved, is still scarce. In order to overcome it and to get a better understanding of the complex regulatory processes; in this study i) a sequence corresponding to the 5’ UTR from the ornithine decarboxylase gene in the red macroalgae Grateloupia imbricata (GiODC) was obtained; ii) in silico analysis of this sequence was performed and DNA regulatory motifs were identified and iii) differential GiODC expression was monitored by RT-qPCR when subjected to different elicitor treatments (i.e. different photoperiods, ethylene and methyl jasmonate). Results show i) the presence of motifs within the 5 'region that are related to studied elicitors and are likely to affect transcriptional expression of GiODC and ii) different GiODC expression patterns according to the time and type of elicitor.
