Neuroprotective effects of thymoquinone-rich fraction nanoemulsion in hypercholesterolemia-induced rat model of sporadic Alzheimer's disease

Hypercholesterolemia is linked to the pathogenesis of sporadic Alzheimer's disease (AD), accompanied with ß-amyloid (Aß) accumulation, oxidative stress, inflammation and memory impairment. This present study investigated the neuroprotective effects of thymoquinone-rich fraction nanoemulsion (TQRFNE) in high fat-cholesterol diet (HFCD)-induced rats. For comparison purposes, thymoquinone nanoemulsion (TQNE) as well as cholesterol lowering drugs (probucol and simvastatin) and AD drug (donepezil) were also studied. Morris water maze test, serum lipid profile and antioxidant capacity, brain lipid peroxidation together with genes and proteins expressions related to antioxidants, inflammation and Aß production were carried out. In comparison to HFCD and other intervention groups, TQRFNE exhibited superior neuroprotective effects by enhancing memory function and serum antioxidant capacity, and reducing serum lipid profile and brain lipid peroxidation. Furthermore, TQRFNE regulates antioxidants, inflammation and Aß production genes and proteins in the rats’ brain. These findings demonstrate for the first time that intervention with TQRFNE plays critical role in regulating such changes. Hence, the present study provides further scientific evidence for the use of TQRFNE as an alternative measures in attenuating neurodegenerative diseases particularly AD.
