Comparison between formalin and alcohol-based fixative solutions on the histology of the rat's cerebellum

Fixative solutions play vital role in the quality of histology slides. We compared the effectiveness of formalin based fixative solution (FB) and alcohol based fixative solution (AB) using coronal slices of adult male healthy Rattus norvegicus cerebelli (n=8). After brain removal, cerebellum was blocked in paraffin before sliced at 5 and 10 micrometers (µ), and then deparaffinated and stained with hematoxyllin-eosin (HE). Slides were observed under light microscope (Nikon, Japan) with 400x of magnification. Images of these slides were taken and then analysed descriptively with Optilab and Image Raster 3 softwares (Indonesia). Overall, the AB groups demonstrated better cellular and cytoskeleton morphology compared to the FB groups. We observed that the morphology of the Purkinje and granule cells in the cerebellar cortices are best demonstrated in the AB5 group. Compared to the FB5, the dendrites of the Purkinje cells are clearer at the AB5 than at the FB5. The neuropils at the granular layer of the AB5 and AB10 groups are nicely striked, compared to those of FB5 and FB10. At the later groups, the dendrites of the Purkinje cells and the neuropils at the granular layer looked mossy and not giving a clear structure of processes like at the AB groups. Furthermore, the nuclei of the granule cells at the FB groups tend to look overstained compared to those of AB’s. Hence, alcohol based fixative solution gave better morphology image compared to the formalin based solution for the rodent’s cerebellum histology in this study.
