Impact of puberty on diabetic nephropathy - Histological evaluation

Introduction. Puberty promotes microvascular complications of diabetes mellitus, including nephropathy. Experimental studies confirm a different renal response to diabetes before and after puberty, probably due to differences in the production of growth factors. Transforming growth factor-β1 (TGF-β1) is a major mediator of diabetic kidney disease. Its renal expression, translation and activation changes with sexual maturation in normal mouse. The role of TGF-β1 in postpubertal susceptibility to diabetic nephropathy was addressed in the present study. Methods. Male Balb/c mice were administered alloxan at 4 weeks of age (weanling) or 12 weeks of age (mature) and treated with insulin to maintain blood glucose levels between 200 and 350 mg/dl. Nonbiabetic controls received saline. After 5 weeks with ad libitum food and water, kidneys were prepared for morphological and immunohistocemical analysis. Results. Morphological analysis showed renal hypertrophy in mature animals compared with the weanling group. Immunohistocemistry for TGF-β1 revealed increased glomerular and renal corpuscles’ staining in mature but not in weanling diabetic mice. Conclusion. Postpubertal male diabetic mice develop renal hypertrophy and stimulation of the renal TGF-β1 system. The above responses are blunted in prepubertal animals. Differences in the regulation of renal TGF-β system may thus explain the postpubertal susceptibility to early manifestations of diabetic nephropathy in male mice.
