Effect of LPS and clomipramine on frequency of epileptic seizures in freely moving WAG/Rij rats

Increase of pro-inflammatory cytokine levels by lipopolysaccharide (LPS) induces alterations in behavior, neurotransmitter function etc. similar to those observed in depressed patients. Depressive illness is associated with activation of the inflammatory response system which could inhibit by tricyclic antidepressants. However, the serotonin and noradrenalin reuptake inhibitor tricyclic antidepressant clomipramine may induce seizure activity. In the present poster we described that LPS induced neuroimmune reactions and clomipramine increased SWD number and duration dose-dependent manner in freely moving WAG/Rij rats. AP5 and LPS showed an enhancing interaction when they were applied in combination. Earlier studies revealed that LPS increase interleukin-1beta level which may increase serotonin and noradrenalin release in the brain. Clomipramine and NMDA-receptor antagonists may cause increase in extracellular monoamine levels. Agonists of 5-HTA1A serotonin receptor and noradrenalin receptor agonist clonidine exacerbated absence seizures. From the results obtained, it appeared that increased level of cytokines, serotonin and noradrenalin may play a role in SWD number and duration increasing effect of LPS and clomipramine.
