Involvement of the Synaptic Adhesion-Like Molecule, SALM1, in learning and memory

The neuronal specific SALM1 (or Lrfn2 - leucine rich repeat and fibronectin type III domain containing 2) belongs to LRRIG (Leucine-Rich Repeat and ImmunoGlobulin) transmembrane protein family that includes the Trk neurotrophin receptor protein. The particularity of SALM1 is that combines a leucine-rich repeat (LRR) and an immunoglobulin-like (Ig) domain. Moreover, this synaptic adhesion molecule contains a PDZ-binding domain (Post synaptic density protein, Drosophila disc large tumor suppressor and zonula occludens-1 protein (amino acids ESTV).
Like many adhesion proteins, SALM1 is expressed early in the developing brain and continue to be expressed in the adult. SALM1 binds both to the NR1 subunit of the NMDA receptor and to PSD-95 (Post Synaptic Density) via this PDZ domain. Reduction of clustering of PSD-95 and NR1 has been observed in SALM1 knockout mice. This adhesion molecule seems to have an essential role in the trafficking of these molecules in order to obtain a functional excitatory synapse.
Interaction with NR1 could be essential for its localisation and stability at the synapse. PSD95 and NR1 are known to be crucial synaptic proteins in learning and memory processes. Therefore, we hypothesized that SALM1 could be crucial in these cognitive functions and set our goal to study the role of SALM1 in learning and memory processes. To up- or down-regulate SALM1, genetic transfer using adeno-associated virus will be used. First, we determined whether training in behavioural tasks could change the expression level of SALM1 and other key synaptic proteins - NR1, PSD-95, and the GluR1 AMPAR subunit.
We show here a traning-specific regulation of the expression of SALM1, NR1, PSD95 and GluR1 in synaptoneurosome preparations following training in two hippocampus-dependent tasks: the contextual fear conditioning and the Morris water maze.
