Sulfated Glycopolymers for glycosaminoglycan mimics and nanomedicine

Glycosaminoglycans (GAGs) are polysaccharides found in living systems that play key roles in biological activities and also function as polyelectrolyte. The preparation of GAGs is important in view of biomaterials development, and total syntheses have been energetically studied by many researchers. However, it is difficult to attain the total syntheses of GAGs due to the complex chemical structure and large molecular sizes.

 We investigated the preparation GAGs mimic polymer and application of nanomedicine via glycopolymer methods. We tried to reconstruct the function of GAGs by glycopolymers of regioselective sulfated saccharides. Glycopolymers provides the strong binding based on the multivalency. In addition, glycopolymer libraries are easily prepared by the radical polymerization. We prepared the phenyl acrylamides with sulfonated N-acetyl glucosamines (GlcNAc) of 6-sulfo-. 3-sulfo-, 4-sulfo and 3,4, 6-sulfo-GlcNAc.  The monomers of sulfonated GlcNAc were synthesized by glycosylation, regioselective sulfonation of GlcNAc and condensation with acrylate.  The monomers were polymerized with other vinyl monomers to prepare the glycopolymer and glyconanoparticles (Figure 1). 

We investigated the affinities of the GAGs mimic glycopolymers with pathogen proteins. The glycopolymers with 6-sulfo-GlcNAc exhibited the inhibitory effect on Alzheimer’s amyloid beta peptides. The inhibitory effects depended on the sugar ratio and the molecular weights of the polymers. The GAGs mimic array was prepared to analyze the interaction with amyloid beta peptides. The enzyme (b-secretase) for Alzheimer disease was also inhibited by the GAGs mimic polymers. The nanoparticle with GAGs mimic monomer was also prepared to capture the specific proteins.

These results showed the usefulness of the glycopolymers comparing to the real GAGs. The glycopolymers are applicable to the many kinds of materials including synthetic polymer, particle, nanoparticles and thin layer formations, which are useful for bionanomaterial and nanomedicine fabrications.
