A novel dental implant material Ti-5Cu with antibacterial and antibiofilm properties

Titanium alloy has been widely used in biomedical applications due to its advantages of excellent corrosion resistance, good biocompatibility and low elastic modulus. As an implant material, however, titanium alloy easily initiates infections after surgery because of its no effect on antibiosis, even though it is one of the most widely used clinical metal materials. In the present study, a new copper-bearing titanium alloy was designed and prepared by proper addition of copper into commercial pure titanium (CP Ti). By using appropriate heat treatment, Ti-Cu alloy has shown good mechanical properties. When contacting with physiological environment, the Ti-Cu alloy has also displayed antibacterial abilities and antibiofilm property via continuously releasing Cu ions during its slight corrosion. For this reason, systematic evaluation and anyalysis on the newly developed Cu-bearing titanium alloy were carried out in this study, including antibacterial and antibiofilmproperties, mechanical properties, corrosion resistance and cytotoxicity. (1) Micro-hardness test, tensile test and electrochemical test were used to evaluate mechanical properties and corrosion resistance of Ti-Cu alloy. The results showed that the addition of Cu improved the mechanical strength. Compared with CP Ti with respect to the electrochemical corrosion resistance, Ti-Cu alloy was not as good as it, but still had good corrosion resistance. (2) Field emission scaning electron microscopy (SEM), fluorescence microscope, confocal laser scaning microscopy (CLSM) and TEM were employed to investigate the antibacterial and antibiofilmeffects of Ti-5Cu alloy. The results showed that the Ti-5Cu could effectively inhibit the formation of biofilm on its surface, and consequently, it behaved excellent abilities to kill Staphylococcus aureus, Escherichia coli, Porphyromonasgingivalisand Streptococcus mutans, which are the commonly seen bacteria in orthopedics and dentistry. (3) Based on the research results, the mechanism of Cu ions acting on the bacteria was tentasively analyzed. The cell wall, cell membrane and DNA of bacteria maightbe destroyed by the Cu ions. On the other hand, the formation of reactive oxygen (ROS) inside the cell is possible to be activated by Cu ions. Due to the deconstruction of cell, the bacteria are evevtually killed. (4) The cytotoxicity of Ti-5Cu alloy was evaluated by using the rat mesenchymal stem cells (rMSCs), and its good biocompatibility was verified.
