As time goes by: a temporal approach to fish genetics

Monitoring changes in genetic diversity as well as their trajectories over time can contribute with critical information for both fisheries and endangered species management. Replication over time allows for the evaluation of the temporal stability of a population and the estimate of its effective size (Ne). This work aimed at testing the temporal model and its potential for Ne estimates in populations with overlapping generations. We compared the inter-annual variation in genetic composition and structure (using mitochondrial and nuclear markers) on species of fish from the Iberian Peninsula. The target species (the shanny Lipophrys pholis, the sandsmelt Atherina presbyter, the white seabream Diplodus sargus and the sea lamprey Petromyzon marinus) were chosen for their contrasting ecological and environmental characteristics. For mtDNA no temporal structure was detected. With the nuclear marker, significant differences were recorded between some sampling periods in the shanny and the sandsmelt. For marine species the haplotype networks revealed deep genealogies, with various levels of diversification, yet with no evident temporal structure. On the other hand, the lamprey presented a very shallow network, also with no temporal structure. L. pholis revealed a smaller Ne/generation when compared to the other species. A. presbyter, D. sargus and P. marinus revealed no evidence of genetic drift for some of the study periods. These results highlight the fact that temporal variations in the composition of the genetic pool should be considered when evaluating the population structure, especially for species with long larval dispersion, thus more vulnerable to recruitment fluctuations.
