In recent years, there is increasing evidence that m6A-RNA methylation plays a key role in the biology of the development progression and therapeutic response of non-small cell lung cancer (NSCLC). m6A-related genes, including METTL3/14, WTAP, FTO, ALKHB5, YTHDF1-3, hnRNPC and eIF3, acted as oncogenes or tumor suppressors were abnormally expressed in the cell lines and tissues of NSCLC. Therefore, some of them are emerging as biomarkers of NSCLC for diagnosis, prognosis, and prediction of therapeutic outcomes, and potential targets for treatment of NSCLC. The flood of novel m6A-related inhibitors has recently provided many dramatic improvements in NSCLC therapy. In this paper, we review the recent research on the biological properties of m6A, the regulatory mechanism of RNA modification and its role in the development of NSCLC, as well as small molecule inhibitors of m6A-modifying enzymes.
