Point-of-Care testing of LMBV infection is crucial to prevent and control disease. Here, we designed a simple and sensitive aptamer-based lateral flow biosensor (LFB) method for the rapid detection of LMBV. In this method, two different aptamers (LA38s and LA13s) specific for LMBV particles were used for target capture and initiation of isothermal strand displacement amplification (SDA), respectively. After adding the reaction product to LFB, the color change of the control zone (C-line) and the test zone (T-line) can be observed by naked eyes within 5 min. The LFB possesses high specificity and accuracy for LMBV detection in vivo and in vitro. The detection limit of LFB is as low as 8 × 101/mL LMBV infected cells, with the detection time of 1 h. Thus, this novel LFB provides a new detection tool, which can be used for rapid detection of LMBV in field.
