DisruptionoftheJEisassociatedwiththedevelopmentofgingivitis,periodontaldisease,and alveolarboneloss.Odontogenicameloblast-associated(ODAM),alsocalledAPINis comprisedofasignalpeptideandanODAMproteinwith12putativeglycosylationsites.It isexpressedduringodontogenesisbymaturationstageameloblastsandisincorporatedinto theenamelmatrixduringtheformationofouterandsurfacelayerenamel.ODAM,asa secretedproteinwhichisaccumulatedattheinterfacebetweenbasallaminaandenamel, mediatestheadhesionoftheJEtothetoothsurface;andisinvolvedwithextracellular signallingofWNTandARHGEF5-RhoA,aswellasintracellularsignallingofBMP-2- BMPR-IB-ODAM.ODAMisalsofoundtobehighlyexpressedinsalivaryglandsand appearstohaveimplicationsfortheregulationofformation,repair,andregenerationofthe JE.Bioinformaticsandresearchdatahaveidentifiedtheanti-cancerpropertiesofODAM, indicatingitspotentialbothasaprognosticbiomarkerandtherapeutictarget.Understanding thebiologyofODAMwillhelptodesigntherapeuticstrategiesforjunctionalepithelium regeneration,dentaldisordersandODAM-relateddiseases.
