Here, we identified a rice lesion mimic mutant lm8015-3, from EMS-induced Zhonghui8015 (ZH8015) library, exhibits orange-yellow spots. The photosynthetic pigments of leaves in lm8015-3 were lower than those in ZH8015 as a result of severely degradation of chloroplast. Detecting of physiological indexes between lm8015-3 and ZH8015 indicated the antioxidation metabolism in the leaves of lm8015-3 were disorganized. The map-based cloning of lm8015-3 revealed a single base substitution (T to A) happened in the first exon of LOC_Os12g16720. lm8015-3 was more susceptible to Magnaporthe oryzae (M.oryzae) compared with ZH8015. The qRT of defense genes between lm8015-3 and ZH8015 indicated lm8015-3 was susceptible to M.oryzae, which was mainly caused by the weakened defense system after inoculation. Besides, we also verified the decreased resistance to M.oryzae of lm8015-3 by leaf-sheath inoculation.
