Many physiological processes and pathological conditions in the human body are regulated by the endogenous molecules adenosine, ATP, and their metabolites. Together with their receptors, membrane channels, and the enzymes that modulate their function and extracellular availability, these components constitute the purinergic system. This system has been extensively studied as a therapeutic target in a wide range of diseases, including cancer, inflammatory disorders, cardiovascular conditions, and neurological pathologies. Notably, several purinergic-targeting drugs have already reached clinical application, highlighting the translational relevance of the purinergic system.

Despite significant advances in the field, the pharmacological potential of the purinergic system remains far from fully explored. This Research Topic aims to highlight recent progress in targeting all components of purinergic signaling, including receptors, enzymes, and membrane channels. We welcome contributions that deepen our understanding of this complex system and its therapeutic implications. Submissions may address a broad spectrum of conditions, with a particular focus on inflammatory diseases, cancer, neurological and psychiatric disorders, cardiovascular conditions, and musculoskeletal pathologies.

In this Research Topic, we aim to bring together recent advances in drug discovery targeting purinergic signaling, spanning from preclinical research to clinical applications. We welcome original research articles and reviews addressing a broad range of topics, including:

• Novel pharmacological mechanisms within purinergic signaling

• Evaluation of endogenous molecules or synthetic analogs as modulators

• Translational aspects bridging basic pharmacology and therapeutic development

• Mechanistic insights into purinergic modulation

• Clinical trials involving purinergic targets

• Innovative methodologies for pharmacological investigation of the purinergic signaling.
