Neurovascular inflammation represents a critical area of research in understanding the dynamics of aneurysm formation and rupture, known precursors to subarachnoid hemorrhage (SAH) and subsequent cerebral vasospasm. These events can lead to secondary neuronal injury, highlighting an important connection to neurodegenerative processes. Pathological neuroinflammation has emerged as a primary driver in these mechanisms, where initial endothelial damage - precipitated by factors like hypertension, genetic predispositions, or lifestyle choices such as smoking - initiates maladaptive vascular inflammation. This cascade involves monocyte infiltration and cytokine release, notably IL-6, which compromises vessel integrity and encourages aneurysmal expansion and rupture.

Recent advances have revealed concentrated inflammatory cell presence in lesion sites and cytokine surges during critical phases, but the intersection of these neurovascular mechanisms with downstream neuronal and neurodegenerative pathways remains underexplored.

This Research Topic will focus on the pathogenic role of neurovascular inflammation in aneurysm progression and its potential implications for neurodegenerative processes, emphasizing mechanistic insights at the molecular and cellular levels. By exploring these pathways, the collection aims to inform strategies that mitigate neuronal injury and clarify underlying neurodegenerative mechanisms.

Dr. Brandon Lucke-Wold is the founder of TauGen, a pharmaceutical company holding patents on ER stress modulators for TBI treatment.
