Background

Pichia pastoris (Komagataella phaffii) stands as a central organism in microbial biotechnology, renowned for its robust capabilities in recombinant protein production, enzyme engineering, and advancements in synthetic biology. Over the last decade, research on this yeast has accelerated, solidifying its status as a preferred platform for both fundamental and industrial applications. Despite major achievements, current challenges remain in optimizing yields, broadening substrate utilization, and engineering strains for more complex biosynthetic tasks. As sustainability and efficiency become more vital in biomanufacturing, the host has emerged as a model system to bridge innovation in basic research with applied solutions in biotechnology. However, concerted efforts are still necessary to fully exploit its potential, address existing bottlenecks, and harness new genetic and systems biology tools.

This Research Topic aims to unite contributions inspired by a conference dedicated to recent developments in Pichia pastoris research, with the objective of highlighting innovative strategies, unresolved obstacles, and visionary applications. By fostering collaboration and showcasing original research, authoritative reviews, and insightful perspectives, the topic will document the strides being made in yeast-based technologies. In particular, it seeks to answer critical questions on how this platform can be further refined as a eukaryotic cell factory, what emerging techniques are on the horizon, and how the organism is driving the next wave of progress in biotechnology.

To gather further insights into the breadth and depth of Pichia pastoris biotechnology, this Research Topic accepts studies exploring both foundational and translational angles, while aligning with the conference's focus. We welcome articles addressing, but not limited to, the following themes:

Novel genetic tools and synthetic biology approaches in Pichia
Strategies for optimizing protein expression and secretion
Systems biology and metabolic engineering in Pichia cell factories
Innovations in fermentation, scale-up, and process engineering
Industrial and pharmaceutical applications of Pichia-based production systems
Emerging trends in sustainability and bioprocess optimization

Case studies linking conference discussions to outstanding questions and future directions.

The collection will accept a variety of article types, including original research, review articles, methods, perspectives, and opinion pieces.

This Research Topic aims to unite contributions inspired by the Pichia2026 conference by welcoming original research and review articles from both conference attendees and the wider Pichia scientific community to address innovative strategies, unresolved obstacles, and visionary applications.
