Limbal stem cell deficiency (LSCD) is a devastating condition that contributes significantly to corneal blindness, particularly in certain regions worldwide, accounting for approximately 15% to 20% of these cases. Limbal stem cells (LSCs), located at the boundary between the corneal and conjunctival epithelium, play a crucial role in the ongoing regeneration of the corneal epithelium. This process is vital for maintaining the transparency and structural integrity of the ocular surface. The loss of LSCs due to various disease processes, such as congenital aniridia, chemical injuries, and autoimmune conditions like Stevens-Johnson syndrome, leads to severe consequences. Patients with LSCD experience persistent epithelial defects, conjunctivalization, and corneal neovascularisation, resulting in chronic pain and significant vision loss. Despite its profound impact, precise global prevalence data are scarce, with estimates largely based on single-centre studies. Recent research emphasises the need for a deeper exploration of LSCD epidemiology and effective treatment pathways.

This Research Topic aims to explore LSCD from multiple perspectives, delving into its epidemiology, pathophysiology, and emerging treatment strategies. The primary objectives include investigating the global prevalence of LSCD, assessing the effectiveness of innovative treatment methods, and testing hypotheses regarding LSC loss and regeneration mechanisms.
