Alzheimer’s Disease (AD) and related dementias affect over 55 million people worldwide and represent a pressing public health challenge. Despite decades of research, the lack of effective therapies that can halt or reverse disease progression remains a critical unmet need. The complex and multifactorial nature of AD, involving genetic, molecular, cellular, and systemic mechanisms, has made it difficult to develop curative treatments.

While early AD therapies focused primarily on symptomatic relief, recent years have seen significant advances in disease-modifying approaches. Monoclonal antibodies targeting amyloid β (Aβ), for example, have begun to show promise in altering disease trajectory. In parallel, progress in advanced therapy medicinal products, including gene, RNA, and cell-based interventions, has opened new avenues for targeting the underlying biology of AD.

Beyond these, emerging therapeutic strategies, such as small molecules targeting tau pathology, anti-inflammatory, and immune-modulating agents, neuroprotective compounds, epigenetic modulators, and repurposed drugs, are also being explored with growing interest. Together, these diverse approaches signal a shift toward more personalized, mechanism-based interventions.

This Research Topic aims to bring together the latest and most promising research efforts aimed at developing innovative therapies for Alzheimer’s Disease and related disorders. We seek to foster a multidisciplinary dialogue that spans molecular research, therapeutic development, and clinical translation.

We welcome Original Research and Review Articles that address, but are not limited to, the following themes:

• New insights into AD pathophysiology and identification of novel therapeutic targets
• Gene-, RNA-, or cell-based therapies for AD
• Emerging pharmacological interventions, including small molecules, immune-modulating agents, and epigenetic therapies
• Repurposed or off-label treatments for AD, including use of existing biopharmaceuticals
• Combination strategies that integrate multiple therapeutic approaches
• Delivery technologies and platforms for innovative therapies
• Clinical trials investigating novel and/or personalized therapies for AD
• Barriers to clinical implementation and equitable access to advanced treatments
