Urethral tissue engineering (UTE) is emerging as a promising approach for the repair and substitution of damaged urethras, addressing debilitating conditions such as urethral strictures. These strictures are prevalent health concerns that can significantly compromise the quality of life and may lead to severe organ damage if left untreated. Over recent decades, while substantial progress has been made in preclinical studies within the UTE field, these advancements have yet to translate adequately into clinical practice. This gap highlights the ongoing need for extensive experimental, preclinical, and clinical studies in UTE.

Recent studies have introduced a range of innovative approaches that are transforming the landscape of UTE. Key advancements include the development of sophisticated biomaterials for scaffold construction, which mimic the urethral microenvironment, enhancing tissue integration and functionality. Likewise, de/re-cellularization techniques have been refined to better preserve and replicate the extracellular matrix, foundational for successful tissue engineering. The use of extracellular vesicles (EVs) has also been identified as a potential game-changer, given their role in cellular communication and regeneration. Moreover, the advent of 3D and 4D bioprinting technologies allows for precise and patient-specific urethral constructs, presenting an exciting frontier for personalized medicine.

This Research Topic aims to position UTE as a viable treatment option in urethral reconstruction. The focus will be on identifying and showcasing novel reports and techniques within UTE, including in vitro studies as well as preclinical animal models. A particular emphasis will be placed on exploring new possibilities and methodologies, such as innovative scaffolds, extracellular matrix (ECM) interactions, de/recellularization procedures, stem cell applications, and the incorporation of advanced bioprinting technologies. Additionally, the scope will highlight the development and application of novel in vitro and animal models that more accurately replicate the complex pathophysiology of urethral disorders, thereby enhancing the translational relevance and predictive value of experimental findings.
