Aging is a complex biological process impacting various organs and systems, prominently affecting the immune system through processes known as immunosenescence and inflammaging. Immunosenescence refers to the gradual decline in immune competence, marked by reduced pathogen defense, lower vaccine response, and a heightened risk of chronic illnesses such as cancer and autoimmune diseases. Inflammaging, a chronic low-level inflammation, further exacerbates tissue degeneration and accelerates aging processes. Recent studies highlight the need to unravel the molecular pathways involved in these phenomena to develop effective therapeutic interventions, improving the healthspan of elderly populations.

This Research Topic aims to dissect the cellular and molecular foundations of immunosenescence and inflammaging, focusing on adaptive and innate immune changes. It seeks to amalgamate insights into biomarker identification, and pioneering therapeutic avenues, including metabolic adjustments, microbial interventions, and novel immune rejuvenation techniques that might hold the potential to alleviate symptoms or reverse aspects of immune aging.

To gather further insights into the boundaries of immunosenescence and inflammaging, we welcome articles addressing, but not limited to, the following themes:

• Age-related alterations in T cell and B cell functionality, including repertoire diversity decline.
• The role of metabolic reprogramming in modulating immune cell functionality during aging.
• Mechanisms of inflammaging, focusing on cytokine imbalance and persistent immune activation.
• Structural changes in lymphoid organs, including the effects of thymic involution and lymphoid tissue remodeling.
• Interactions between the gut microbiota and the immune system in aging, and their impact on overall immune balance.
• Development of novel biomarkers and therapeutic strategies to counter immune aging.

We encourage submissions of original research articles, reviews, mini-reviews, and perspective papers offering fresh perspectives on these critical aspects of immune aging and therapeutic strategies. Contributions from clinical studies, animal model investigations, or computational analyses are especially welcomed to enrich this multidisciplinary dialogue.
