Bone disorders, including osteoporosis, arthritis, fracture non-union, bone cancer and metastasis, and other rare bone disorders, represent a significant global health burden. These conditions are characterized by impaired bone tissue repair and remodeling processes, leading to structural compromise and functional decline. A deeper understanding of the molecular mechanisms governing bone regeneration and remodeling, coupled with the development of targeted therapeutic strategies, is crucial for improving patient outcomes and quality of life. This research topic aims to highlight the latest advances in basic and translational research focused on bone tissue repair and remodeling. Clinical studies, patient samples, and any research involving patient participation are strictly out of scope for this Research Topic.

This Research Topic aims to collect cutting-edge research and comprehensive reviews that address the complex mechanisms underlying tissue repair and remodeling in bone disorders. The role of key signaling pathways, cellular interactions, and extracellular matrix components in regulating bone regeneration and remodeling will be explored. The special issue also seeks to showcase innovative therapeutic approaches that target these molecular mechanisms to promote bone repair, prevent disease progression, and restore skeletal function. It is highly recommended that article submissions perform in-depth signalling studies, multi-omics, high-throughput screening, translational medicine and precision medicine to achieve better understanding and treatment of bone diseases and associated complications.

This Research Topic welcomes Original Research Articles, Reviews, Mini Reviews, Technical reports, Short communications, and Perspective articles may include, but are not limited to:
1) Molecular Mechanisms of Bone Repair and Remodeling: Signaling pathways (e.g., Wnt, BMP, TGF-β, RANKL), transcription factors, and epigenetic regulators involved in osteoblast and osteoclast differentiation, activity, and bone matrix synthesis.
2) Cellular Interactions in Bone Regeneration: Studies on the cellular interactions and role of osteoblast, osteoclast, osteocytes, immune cells, endothelial cells, and mesenchymal stem cells (MSCs) in bone repair and remodeling processes.
3) Extracellular Matrix and Biomaterials: Basic and translational Research on the composition, structure, and mechanical properties of the bone matrix and their influence on bone cell behavior.
4) Targeted Therapies for Bone Disorders: Basic and translational Research on development and evaluation of therapeutic strategies targeting specific molecular pathways or cellular processes involved in bone disease, including small molecules, biologics, gene therapies, and cell-based therapies. All studies must be preclinical and should not involve patients or clinical data.
5) Advanced Imaging and Diagnostic Techniques: Basic and translational Research on application of advanced imaging modalities for monitoring bone repair and remodeling in vivo and for assessing the efficacy of therapeutic interventions. Only preclinical studies, excluding those involving patient data or clinical subjects, are within scope.

Manuscripts that provide basic, mechanistic and translational insights into bone biology, explore novel therapeutic targets, or report on the translation of innovative therapies for bone disorders are of particular interest. Studies that are clinical or purely epidemiological, including those that identify risk factors or develop predictive models for bone diseases, are not within the scope of this research topic. Clinical Trials are not accepted in this journal, under any article type that Frontiers in Bioengineering and Biotechnology offers.
