Intranasal (IN) drug delivery is a safe, non-invasive method that offers significant advantages for the treatment of neurological and other central nervous system (CNS) diseases. This route enables both direct transport of therapeutics from the nasal cavity to the brain via olfactory and trigeminal pathways, effectively bypassing the blood-brain barrier, and indirect absorption into the systemic circulation. IN administration allows for rapid onset of action, reduced systemic side effects, and improved patient compliance compared to oral or intravenous routes.

It is particularly promising for delivering neuropeptides, monoclonal antibodies, and gene therapies, which often face challenges in crossing the blood-brain barrier. Recent research highlights the potential of IN delivery to enhance the efficacy and safety of CNS treatments. However, optimizing drug formulations and delivery devices remains essential to ensure consistent dosing and maximize therapeutic outcomes.

The goal of this Research Topic is to provide a comprehensive platform of Original Research, Review, and Perspective articles that collectively advance the field of nasal drug delivery systems with a particular focus on innovative nanoformulations, for treating CNS disorders. Additionally, it seeks to outline future directions for clinical translation, regulatory considerations, and interdisciplinary collaborations that will accelerate the development of nasal medications.

We welcome the submission of manuscripts including, but not limited to, the following topics. Special focus will be given (but is not restricted) to:

• Drugs used or potentially suitable for CNS disorders that may, due to physicochemical and pharmacokinetic challenges, benefit from nasal administration.

• Polymers used or potentially suitable for nasal nanoformulations.

• Nasal formulations designed for the treatment of CNS disorders.

• Regulatory considerations related to nasal formulations, with a particular focus on nanoformulations.

• Devices used or potentially suitable for targeted deposition of nanoformulations in the olfactory region.
