This is a Research Topic initiated by ISACB, the International Society for Applied Cardiovascular Biology.

Aortic stenosis, largely driven by calcific aortic valve disease (CAVD), remains one of the most prevalent and clinically significant valvular pathologies in aging populations. Despite advancements in surgical and transcatheter valve interventions, no effective medical therapies are currently available to halt or reverse the progression of valvular calcification. This Research Topic aims to spotlight the latest innovations and conceptual advances in the field of CAVD by exploring the cellular, molecular, biomechanical, and systemic mechanisms underpinning disease initiation and progression, and by discussing innovative clinical procedures.

We seek to foster interdisciplinary dialogue and promote translational breakthroughs that bridge basic research with clinical applications. Particular emphasis will be placed on novel mechanisms of endothelial dysfunction, inflammation, lipid mediators, metabolic drivers, extracellular matrix remodeling, and fibrocalcific transformation, as well as on innovative technologies such as advanced imaging, omics approaches, mechanobiological modeling, organoid development, and tissue-engineered platforms. We will also focus on emerging surgical procedures. The role of genetic and epigenetic regulation, as well as emerging therapeutic targets and device strategies, will also be considered.
