The immune system’s ability to mount an anti-tumor has long been recognized, and strategies to leverage the immune system to treat cancer were subsequently developed. In recent years, therapies that redirect T cells to target malignant cells such as chimeric antigen receptor T-cells (CAR-T) and bispecific T-cell engaging molecules have proven very successful, particularly in B cell and plasma cell disorders. However, treatment failure and toxicity remain significant concerns with currently available T-cell redirecting therapies, and the development of these treatments for myeloid malignancies and solid tumors has been slow. Ongoing research to optimize the use of current T-cell redirecting therapies and develop more effective agents across a wide range of tumor types is needed.

The goals of this Research Topic are as follows:

● Review the current state of T-cell redirecting therapies in cancer care, highlight new indications and combination therapies using the currently available agents.

● Highlight current research involving toxicity management, predicting treatment failure, strategies to prevent relapse, and use in specific patient populations.

● Review emerging cellular therapies and T-cell engagers in both solid tumors and hematologic malignancies.

● Review ongoing challenges in immunotherapy and cellular therapy development for specific tumor types.

The research topic includes current and emerging T-cell redirecting therapies in both solid tumors and hematologic malignancies. Within hematologic malignancies, articles focusing on lymphoma and myeloid malignancies are of specific interest as multiple myeloma was the topic of a recent edition of Frontiers Immunology. Therapies of interest include CAR-T, bispecific T-cell engagers (BiTEs), and bispecific or multi-targeted antibodies. Treatments using other immune effector cells (i.e., NK cells) and allogeneic cells are of interest, as are novel approaches to cell therapy manufacturing such as in vivo CAR-T manufacturing.

We are interested in the following types of manuscripts:

● Review articles
● Original research
○ Retrospective/Case Series/Outcomes research
○ Translational research pertaining to mechanisms of resistance to current T-cell redirecting therapies, toxicity mitigation, manufacturing, novel targets
● Case reports highlighting unique treatment approaches or rare toxicities

Please note that Dr Jason Romancik is part of the advisory boards with KITE, Bristol Myers, Squibb, Astra Zeneca, and ADC. Also note that manuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases which are not accompanied by robust and relevant validation (clinical cohort or biological validation in vitro or in vivo) are out of scope for this Research Topic.
