Advances in genome editing have transformed the development of animal models used to understand human diseases. While transgenic mouse models have broadly contributed to many insights of disease mechanisms, their translational limitations, such as small size, different physiology and organ structure, can make it challenging to fully represent human disease progression and treatment outcomes. As a result, large animal models (e.g., pigs, sheep, and goats) are increasingly recognized as better alternatives due to their closer anatomical and physiological resemblance to humans.

Recent advancements in CRISPR/Cas9-based genome editing technologies have revolutionized the scope of genetic modulation in animal research. This technology allows researchers to introduce precise and human-derived mutations into strategic genes, thereby replicating disease phenotypes observed in humans. CRISPR/Cas9 not only enhances the fidelity of genetic alterations but also optimizes the creation of more representative animal models. This progression is pivotal for deciphering gene regulation and underlying molecular mechanisms in various diseases, in addition to providing invaluable platforms for testing new therapies and evaluating their molecular impacts comprehensively.

This Research Topic focuses on innovations that advance the development and refinement of large animal models associated with various human diseases. It also seeks contributions to genomic modifications intended for the well-being and research related to these conditions.
