Mitochondrial respiratory complexes are essential enzymes located within the inner mitochondrial membrane and play a critical role in the electron transport chain (ETC). This vital pathway includes Complexes I through IV, which facilitate electron flow and drive proton pumping to create an electrochemical gradient. This gradient is harnessed by Complex V (ATP synthase) for the production of ATP. Although substantial progress has been made in understanding the general functions of these complexes, detailed knowledge of their structural organization, molecular interactions, and dynamic functions in their natural environments remains incomplete. Recent advancements in structural biology—particularly in cryo-electron microscopy (Cryo-EM) and cryo-electron tomography (Cryo-ET)—have significantly enhanced our ability to capture high-resolution images and 3D reconstructions of mitochondrial respiratory complexes in near-physiological conditions.

This research topic aims to compile and present the latest insights obtained using Cryo-EM and Cryo-ET to explore mitochondrial respiratory complexes at molecular and cellular levels. Specifically, the research will provide structural insights into the arrangement of individual complexes, their interactions with neighboring complexes or other mitochondrial components, their conformational changes during function, and how structural disruptions may lead to dysfunction and disease. This research will address unresolved questions related to electron transfer mechanisms, proton pumping efficiencies, structural dynamics in response to metabolic changes, and possible therapeutic interventions for mitochondrial disorders.
