ABC transporters are involved in a variety of physiological functions, and their dysfunction can lead to the development of various diseases such as cystic fibrosis, stargardt disease, X-linked adrenoleukodystrophy, and sitosterolemia; elucidating the molecular mechanisms of ABC transporter dysfunction is crucial for developing therapeutic strategies. In addition, modulation of biosynthesis and function of transporters by small molecule compounds such as pharmacological chaperones and proteostasis regulators is an attractive approach to cure pathologies.
Recent technological advances, such as ligand-based and structure-based virtual screening combined with MD simulation, facilitate the identification of chemicals that bind ABC transporters and may affect their biosynthesis and function. Understanding the mechanisms of action of chemicals that affect ABC transporters is important for understanding pathomechanisms and for designing combination approaches to maximize the therapeutic effects of chemicals. The ultimate goal of this topic section is to lay the foundation necessary to develop new therapeutic strategies for diseases associated with the dysregulation of ABC transporters.
