APOE4 is the primary genetic risk factor for the sporadic, late-onset form of AD. However, very little is understood about how the pathogenesis of AD differs in APOE4 carriers vs. non-carriers. The association between APOE4 and increased risk of AD is well established, but there is also evidence to suggest that APOE4 carriers vs. non-carriers differ in the pathological and cognitive presentations of the disease itself. There are also open questions about how environmental and genetic factors differentially impact the AD susceptibility between APOE4 carriers and non-carriers, the elucidation of which will have important implications for the prevention and treatment of AD.
The goal of this Research Topic is to publish novel research articles that help to elucidate the heterogeneity in both the susceptibility to and presentation of AD between APOE4 carriers and non-carriers. How do MCI or AD patients who are APOE4 carriers vs. non-carriers differ in their physiological, pathological, or cognitive traits? Do specific factors differentially affect the risk of cognitively normal APOE4 carriers vs. non-carriers eventually developing AD? Are there potential therapeutic interventions against AD that are more effective in APOE4 carriers vs. non-carriers, or vice versa?
The primary focus of this Research Topic will be studies that utilize human data from middle-aged or elderly individuals in order to elucidate differences in the susceptibility to or presentation of AD between APOE4 carriers vs. non-carriers. This can include postmortem tissue data, neuropsychological data, imaging data, therapeutic data, or any other relevant human data. Studies that specifically investigate or include data about race/ethnicity differences or sex differences in APOE4-associated heterogeneity are strongly encouraged. Mouse or cell-culture studies will not specifically be excluded from this Research Topic, but these studies must directly relate to how APOE4-associated heterogeneity impacts the development or presentation of AD.
