Background

Tumor immune evasion remains one of the greatest obstacles to achieving durable responses in cancer therapy. Tumors often employ multiple and overlapping mechanisms to escape immune recognition and destruction, such as downregulation of antigen presentation, modulation of immune checkpoints, and alteration of the tumor microenvironment. With advances in genome editing technologies—including CRISPR/Cas9, base editing, and prime editing—researchers now have powerful tools to simultaneously target and modify multiple genes, enabling a new generation of precision immunotherapies.

This Research Topic aims to showcase the latest developments and applications in multiplex genome editing designed to overcome tumor immune evasion.
