Background

Autoimmune diseases (AIDs) and major chronic non-communicable diseases (NCDs), including cardiovascular diseases, diabetes, chronic respiratory diseases, and cancer, represent significant global health burdens with complex, multifactorial etiologies. Despite advances in understanding their pathogenesis, the interplay between genetic predisposition, environmental triggers, and molecular dysregulation remains poorly defined. Recent progress in multi-omics technologies—genomics, transcriptomics, proteomics, metabolomics, and epigenomics—has opened unprecedented opportunities to unravel shared mechanisms and heterogeneity across these conditions. For instance, immune dysregulation, chronic inflammation, and metabolic disturbances are common threads linking AIDs and NCDs, yet their molecular underpinnings are underexplored. Integrating multi-omics data holds promise for identifying novel biomarkers, therapeutic targets, and personalized intervention strategies. However, challenges persist in data integration, functional validation, and translating findings into clinical practice. This Research Topic aims to bridge these gaps by fostering interdisciplinary collaboration and advancing innovative methodologies to decode the complex molecular landscapes of AIDs and NCDs.
This Research Topic seeks to address the critical need for a holistic understanding of autoimmune diseases and major chronic NCDs through multi-omics approaches. By leveraging cutting-edge technologies and computational tools, we aim to uncover shared molecular pathways, disease-specific signatures, and environmental interactions driving these conditions. Recent advances in single-cell sequencing, spatial omics, and AI-driven data integration offer transformative potential to dissect disease heterogeneity and progression. We encourage submissions that propose novel frameworks for multi-omics data harmonization, mechanistic insights into immune-metabolic crosstalk, and translational applications for precision medicine. Our goal is to catalyze breakthroughs that enhance diagnostics, therapeutics, and preventive strategies for these globally prevalent diseases.

We invite original research, reviews, methods, and perspective articles addressing, but not limited to, the following themes:
• Multi-omics integration: Novel computational or experimental approaches to harmonize genomic, transcriptomic, proteomic, and metabolomic datasets.
• Mechanistic insights: Shared pathways (e.g., inflammation, autophagy) and cross-disease comparisons between AIDs and NCDs.
• Biomarker discovery: Identification of diagnostic, prognostic, or predictive biomarkers using multi-omics platforms.
• Translational applications: AI/ML-driven tools for drug repurposing, personalized therapy, or risk stratification.
• Environmental interactions: Role of microbiome, diet, or pollutants in modulating disease trajectories.
• Ethical and clinical challenges: Data privacy, equity in omics research, and barriers to clinical implementation.
