Background

Alzheimer’s disease (AD) is the most common neurodegenerative disorder affecting older adults worldwide, yet effective disease-modifying therapies are yet to be achieved. Amyloid-beta (Aβ) has been predominant in the therapeutic landscape, driven by hypotheses linking its accumulation to the pathogenesis and progression of Alzheimer’s disease. Recent years have seen the clinical approval of several Aβ-targeting monoclonal antibodies, generating interest and controversies around their efficacy, safety, and real-world applications. Simultaneously, the field is experiencing growing interest in alternative approaches, including tau-targeted therapies, neuroinflammation modulation, neuroprotection by modulation of brain energy metabolism, and lifestyle interventions, reflecting the complex nature of AD.

This Research Topic aims to provide a platform to investigate the current state and future directions of Alzheimer’s disease therapeutics. We invite research exploring both the potential and challenges of amyloid-beta-directed interventions, as well as innovative therapeutic strategies and targets beyond amyloid. By investigating diverse approaches, this collection seeks to move the field toward effective therapeutic solutions for AD.

We welcome submissions in the form of original research, reviews, perspectives, and commentaries on topics including, but not limited to:

• Efficacy, safety, and long-term outcomes of amyloid-beta therapies

• Insights into amyloid-beta pathology and intervention

• Targeting neuroinflammation, oxidative stress, and synaptic dysfunction in AD

• Combination or multi-targeted therapeutic strategies

• Biomarkers for predicting and monitoring treatment responses

• Repurposing drugs and novel small molecule approaches
