Development of antibody-cocktail for simultaneous detection of ASGV, ACLSV and ASPV

Production of apple at both quantitative and qualitative level is hampered on a large scale due to multiple viral infection causing huge economic losses. Mixed infection of apple stem grooving virus(ASGV), apple stem pitting virus(ASPV) and apple chlorotic leaf spot virus(ACLSV) are common in apple trees. Diagnosis of viruses by serological techniques requires development of antibodies against each virus individually (mostly exported in India), which is not only costly but also time-consuming. Therefore, we designed a triple construct gene for expressing a recombinant protein constituting conserved region in coat protein(CP) of ASGV, ASPV and ACLSV, for the development of an antibodycocktail against three apple viruses for their simultaneous detection in a limited time. We have tried to develop an antibody-cocktail that can perform diagnostics for three different apple viruses in a single ELISA assay, that would not only be time-saving but also cost-effective for the purpose of testing large samples during certification of plants in nurseries. To our knowledge, this is the first report of diagnosis of multiple viruses of apple in a single ELISA assay by using antibody-cocktail.
