Background

The ongoing challenge posed by viral infections to global health has been exemplified by the SARS-CoV-2 pandemic and the continuous threats from pathogens such as influenza and HIV. Despite advancements in antiviral therapies, there remains a critical need for novel strategies that harness the potential of antibody-based therapeutics. Known for their high specificity, versatility, and reduced off-target effects, antibodies have emerged as a formidable strategy against viral infections. Landmark approvals, such as palivizumab for RSV, tixagevimab/cilgavimab, and sotrovimab for SARS-CoV-2, and ibalizumab for multi-drug resistant HIV, underscore their clinical impact. Recent advancements in antibody engineering, including bispecific constructs, nanobodies, and antibody-drug conjugates, have expanded their utility, offering hope for better pandemic preparedness and targeted treatment interventions.

This Research Topic aims to delve into the innovative strides made in antibody-based therapeutics specifically targeting viral infections. Our objective is to explore novel engineering methodologies, underlying mechanisms of action, and improved delivery strategies, thereby accelerating the integration of these biologics into clinical practice. By highlighting key developments in antibody design and deployment, we seek to strengthen the therapeutic arsenal available for established and emerging viral threats, enhancing overall preparedness for future pandemics.

To gather further insights into the potential and challenges of these therapies, we invite articles exploring antibody-based strategies with a focus on:

- Engineering next-generation antibody formats for targeting viral pathogens
- Implementing Fc engineering to enhance antibody performance in antivirals
- Leveraging novel antibody discovery platforms to identify neutralizing antibodies
- Developing antibody cocktails and combination therapies for enhanced efficacy
- Clinical applications and case studies of engineered antibodies against viral infections
