Background

Adjuvants are critical components in vaccine formulations, enhancing their immunogenicity and protective efficacy. Over the years, a variety of adjuvants have been approved for use in both animals and humans, with ongoing research and development efforts worldwide. Despite these advancements, there remains a pressing need to develop vaccines for diseases that currently lack them, including those not yet in clinical trials. Additionally, understanding the selection of appropriate adjuvants and the immune molecular mechanisms they trigger is crucial. The development of specific adjuvants for parenteral or mucosal vaccines presents unique challenges, necessitating transdisciplinary approaches that have led to innovative adjuvant prototypes. Recent advancements in adjuvant research include in silico design, biological and chemical origins, and novel methods of chemical synthesis, all contributing to the broad integrated knowledge that continues to benefit humankind. However, gaps remain in understanding the best routes for administration and the mechanisms of action of these adjuvants, highlighting the need for further investigation.

This research topic aims to integrate and discuss emerging advances in the research and development of novel adjuvants, as well as to highlight the expanding knowledge in the context of immunity and vaccinology for the prevention, control, and treatment of diseases affecting both animals and humans. The objective is to offer a platform for novel, revolutionary, and cutting-edge scientific findings on adjuvants, focusing on their design, origin, type, and efficacy. By analyzing the knowledge obtained so far and identifying gaps in adjuvants in authorized vaccines, as well as those in clinical and non-clinical trials, this research seeks to improve the understanding of adjuvant mechanisms of action and enhance the potency and effectiveness of vaccines.
