Drug resistance remains one of the most formidable challenges in modern oncology. While significant advances in cancer diagnosis and treatment have improved patient outcomes, many cancers eventually recur and become unresponsive to standard therapies. A growing body of research suggests that a subpopulation of tumor cells, cancer stem cells (CSCs), is critical in driving this resistance. These cells are distinguished by their capacity for self-renewal, differentiation, and remarkable resilience to conventional therapies. As such, cancer stem cells have become a focal point for researchers seeking to understand and overcome the mechanisms underlying therapeutic failure and disease relapse.
CSCs contribute to tumor heterogeneity, metastatic potential, and treatment resistance through diverse mechanisms, including enhanced DNA repair capabilities, activation of anti-apoptotic pathways, drug efflux via ABC transporters, and metabolic reprogramming. Their ability to enter a quiescent or dormant state further allows them to evade therapies that target rapidly dividing cells. Understanding these survival strategies is key to developing more effective, durable treatments for cancer patients.
This Research Topic, "Unlocking Cancer’s Secrets: Overcoming Drug Resistance in Cancer Stem Cells," aims to consolidate and expand our knowledge of CSC biology and resistance mechanisms. We invite researchers to contribute studies that explore the underlying molecular and cellular processes that enable CSCs to withstand therapeutic assault and investigate their role in cancer progression, recurrence, and metastasis. A particular emphasis will be placed on identifying novel therapeutic targets and strategies capable of eliminating CSCs or sensitizing them to existing treatments.
We are especially interested in submissions investigating the potential of natural compounds and bioactive plant extracts in targeting CSC-associated resistance pathways. Nature remains a rich source of pharmacologically active substances, and many traditional medicines have shown promise in preclinical models. Furthermore, identifying and validating biomarkers predictive of CSC-driven resistance may open new avenues for personalized medicine, allowing clinicians to tailor therapies based on a tumor’s stem-like cell content and resistance profile.
To this end, we welcome original research articles, comprehensive reviews, and thought-provoking perspectives or opinion pieces that shed light on this critical aspect of cancer biology. Topics of interest include, but are not limited to:
