Background

As a leading Open Access publisher, Frontiers is committed to empowering not only scientists, but other researchers, innovators and members of the public. As such, highlighting recent advances and the real-world applications of Predictive Toxicology are key parts to the agenda of Frontiers in Pharmacology. Predictive toxicology uses and integrates in silico, in chemico and in vitro approaches named “New Approach Methodologies (NAMs)” to predict the potential toxicity and toxic potency of chemicals or drugs in humans. Predictive Toxicology is moving beyond the realm of drug development and now forms the backbone of next generation risk assessment (NGRA) for chemical risk assessment, which implements NAMs to assure human safety without performing animal testing. This Research Topic in Frontiers of Pharmacology aims to highlight the latest advancements in the techniques, policies, materials, and methods used to develop and test products for the market.



This Research Topic encourages contributions from scientists describing their insights and forecasts on the ways in which Predictive Toxicology can be utilised in the pharmaceutical, agrochemical, cosmetic, and chemical industries, and for regulatory decision making. Predictive toxicology applications for selection of first and safe dose in man and prediction of side effects of drugs/chemicals are of interest.

• Current development of toxicokinetics (TK) and toxicodynamic (TD) assessment.

• PBPK modeling and QIVIVE in chemical/drug safety assessments.

• Opportunities and challenges of integrating artificial intelligence (AI) approaches into TK assessments.

• Microphysiological systems (organ-on-a-chip (MOC))

• In silico computational toxicology.

• Success and failures using different first-in-human (FIH) pharmacokinetic prediction approaches
