Background

The field of chemical biology has made significant strides in improving our understanding and treatment methodologies for bone and bone marrow diseases, including orthopedic and hematological diseases, such as osteoporosis, osteoarthritis, osteolysis, spinal diseases, anemia, multiple myeloma, and various leukemias. These conditions impose substantial health burdens globally, with currently available medical treatments often falling short due to limited efficacy and adverse side effects. Cutting-edge research in chemical synthesis and medicinal chemistry has opened new avenues for the creation of potent compounds that hold therapeutic promise in tackling these diseases. However, the pathway from chemical discovery to effective clinical application presents numerous challenges, necessitating a focused exploration of novel compounds' chemistry and their translational potential.

This Research Topic seeks to harness the analytical and synthetic capabilities central to chemical research to identify, design, and validate bioactive compounds targeting bone and bone marrow pathologies. Key objectives include evaluating these compounds' preclinical efficacy through robust chemistry-led approaches, exploring their safety and mechanism of action, and facilitating their transition into early-phase clinical trials. Specific inquiry areas encompass promising chemical structures emerging from preclinical studies, the translation of these findings into clinical settings, and the elucidation of their functional pathways at the molecular level. Meanwhile, the retrospective clinical research on the application of new drugs applied in clinical is also welcomed.

To gather further insights within the bounds of chemistry-driven research for bone and marrow therapies, we invite articles addressing, but not limited to, the following themes:

• Mechanistic studies of compound interactions at the molecular chemistry level
• Innovative analytical methods for evaluating compound efficacy and safety
• Chemistry-tied pathways facilitating the clinical translation of novel treatments
• Preclinical research of novel chemically synthesized or naturally isolated compounds on novel disease models
• Clinical trial design and outcomes (randomized controlled trial and retrospective research) for compounds targeting bone and marrow conditions
• Mechanistic studies unraveling molecular pathways of novel therapies
