Genome-Wide Identification and Analysis of OSC Gene Family in Panax japonicus

Panax japonicus is an important medicinal plant rich in triterpenoids. However, the biosynthetic pathway of triterpenoids in remains poorly understood. In this study, we employed a genome-wide strategy to systematically identify twenty-one candidate 2,3-oxidosqualene cyclases in P. japonicus (PjOSC) genes. Meanwhile, the conserved patterns of promoters, gene structures and cis-acting elements were also analyzed. Moreover, these PjOSC genes were distributed on 11 chromosomes. We conducted a synteny analysis to understand the evolution of the P. japonicus PjOSC gene family. A total of 19 gene pairs of PjOSCs were identified through tandem or segmental duplication events. Promoter prediction revealed the presence of various cis-acting elements related to abiotic stresses and phytohormone response within the 2 kb upstream region of PjOSCs. Transcriptome and qRT-PCR analyses demonstrated that the expression of PjOSCs is tissue-specific. Collectively, these findings suggest that PjOSCs play a key role in the synthesis of triterpenoids in P. japonicus. The results provide an understanding of PjOSCs in P. japonicus and lay the foundation for a further functional analysis of PjOSC genes in the triterpenoid anabolism of P. japonicus.
